Background: Urinary tract infection (UTI) is one of the most common bacterial infections and may be caused by Pseudomonas aeruginosa. This bacterium is capable of forming biofilms, which contribute to bacterial persistence and antibiotic resistance. The Polysaccharide synthesis locus D (pslD) gene plays an important role in biofilm formation and can be used as a molecular marker for the detection of biofilm-producing bacteria.

Purpose: To detect the pslD gene in urine samples from UTI patients using the Polymerase Chain Reaction (PCR) method.

Method: This descriptive study used 10 urine samples from UTI patients and 2 Pseudomonas aeruginosa isolates as positive controls. DNA was isolated using the Presto™ Mini gDNA Bacteria Kit, followed by qualitative and quantitative tests, annealing temperature optimization, PCR amplification, and visualization using agarose gel electrophoresis.

Results: Qualitative analysis confirmed the presence of DNA in all samples. The optimum annealing temperature was determined to be 63°C. PCR amplification produced DNA bands around the target size of 369 bp in positive controls and several urine samples. However, smears, non-specific bands, and variations in band intensity were observed among samples. These variations may be associated with DNA quality, the presence of urinary inhibitors, and differences in the causative microorganisms.

Conclusion: The pslD gene could be detected using PCR and may serve as a molecular marker associated with biofilm-forming bacteria in UTI patients.
